Abstract

Rimpang temulawak mengandung senyawa metabolit sekunder berupa flavonoid, steroid, triterpenoid dan tannin. Penelitian ini bertujuan untuk mengetahui efek gastroprotektif ekstrak etanol rimpang temulawak melalui parameter makroskopik lambung, volume, pH, keasaman total cairan lambung, kadar mucin, serta pengamatan mikroskopik. Tikus dibagi menjadi 6 kelompok yaitu kelompok normal, kontrol positif (Omeprazole 20mg/kgBB), kontrol negatif (NaCMC 0,5%), kelompok 1 (Ekstrak 100 mg/kgBB), kelompok 2 (Ekstrak 200 mg/kgBB), dan kelompok 3 (Ekstrak 400 mg/kgBB). Tikus diberi perlakuan selama 14 hari, kemudian diinduksi etanol absolut pada hari ke-15. Kadar total flavonoid, kurkuminoid, dan fenolik yang terkandung dalam ekstrak rimpang temulawak masing-masing sebesar 49,962 mgQE/g, 124,51 mgCE/g dan 108,234 mgGAE/g. Hasil penelitian menunjukkan terdapat perbedaan lesi mukosa lambung, volume, pH, keasaman total cairan lambung, serta kadar mucin yang signifikan dengan kelompok kontrol negatif (p<0.05) serta perbedaan yang tidak signifikan pada kelompok 3 dengan kelompok kontrol positif (p>0.05) yang menandakan bahwa ekstrak etanol rimpang temulawak mampu memberikan perlindungan terhadap lambung tikus. Dosis 400mg/kgBB paling efektif memberikan efek gastroproteksi mendekati kontrol positif pemberian Omeprazole 20 mg/kgBB. Semakin tinggi dosis ekstrak yang diberikan, maka semakin baik efek gastroproteksi yang dihasilkan.
