Abstract

Hyperuricemia is a condition that elimination of the uric acid had been disruption through the kidneys and will effect to decreased uric acid secretion. So that there will be an increase of uric acid levels in blood.One of the plants that can be used to reduce uric acid levels is the leaves of Ruku-ruku (Ocimum tenuiflorum Linn.). Ruku-ruku leaves are known to contain flavonoid compounds which have the potential to reduce uric acid levels in the blood. Flavonoidcompounds apigenin and luteolin in the leaves of Ruku-ruku can work as competitive inhibitors of the xanthine oxidase enzyme to inhibit the formation of uric acid in the kidneys. This study aims to determine the antihyperuricemia effect of Ruku-ruku ethanol extract on male white rats wistar strain induced by chicken liver juice and potassium oxonate. A total of 25 wistar strain rats aged 2-3 months with a weight range of 150-250 grams were divided into five groups: positive control (allopurinol 1.96 mg / 200 gBB), negative control (0.5% Na CMC), group I ( 2.25 gram / kgBB), group II (4.5 gram / kgBB), and group III (9gram / kgBB). The whole group was induced by oral chicken liver juice every day for 3 days at a dose of 3mL / 200 gBB and potassium oxonate on day 4 250mg / kgBB intraperitoneallyto achieve hyperuricemia condition, then it was given a test preparation for 8 days after induction. The results showed that PKAU% in groups I, II, and III were 44.22%, 53.11%, and 71.71%. In group III it has the best antihyperuricemia activity because the decrease is almost close to allopurinol 72.63%. The analysis results obtained showed a decrease in uric acid levels and there were significant differences compared to the negative group (p <0.05). Based on the relationship of the percentage decrease in blood uric acid levels to the dose, it was found that ED50 ethanol extract of Ruku-ruku leaves was 3,93 gram / KgBB.
