Abstract
A study of the effect of glibenclamide loaded solid lipid nanoparticle (SLN) on decreased blood glucose levels was imposed on normal rats. Glibenclamide loaded SLN production of 3 batches with each EE percentage of 83.023 ± 1.725%, 81.536 ± 0.664%; and 81.903 ± 0.953%. The resulting glibenclamide loaded SLN preparation has an average particle size of 67.1 nm, PDI 0.061, and zeta potential -1.4 mV. Glibenclamide loaded SLN had a greater diffussion percentage at 720 minutes than tablet of glibenclamide on the market and glibenclamide pure powder, 36,46 ± 0,535%, 33,563 ± 0,271%, dan 24,140 ± 0,146%, respectively. This study used the glucose tolerance test, by way of burdening the normal rats that have been fasted for 20 hours with glucose. Body weight of rats used 150 – 250 g, age 2 – 3 months. Rats were divided into five groups, including negative control group (Na CMC 1%), positive control group (tablet of glibenklamid dose 1.89 mg/kgBB), glibenclamide loaded SLN group with three dosage ratings 0.95; 1.89; and 3.78 mg/kgBB. Giving glucose at loading is 6.75 g/kgBB. Blood was taken from the lateral veins of the tail at minute 0, 30, 60, 90, 120, 150, and 180. Blood glucose levels determined using a EasyTouch® glucometer. AUC0-180 of each group were 26043 ± 33.63 (negative control), 17340 ± 10.39 (positive control), 16587 ± 11.73 (dose 1), 15618 ± 12.96 (dose 2), and 14700 ± 17.00 (dose 3). Percent reduction in blood glucose levels obtained by positive control with dose of 1.89 mg/kgBB in the amount of 50,190%, while glibenclamide loaded SLN at a dose of 0.95; 1.89; and 3.78 mg/kgBB showed percent reduction in blood glucose levels of 57.008%; 66.749%; and 77.163%. Based on statistical test using one way ANOVA with 95% confidence level, there were no significant differences between group of positive control with group of dose 1, dose 2, and dose 3 (p>0.05).
