Abstract

Noni fruit (Morinda citrifolia L.) contains flavonoids that have immunomodulatory activity. Orally Dissolving Film (ODF) formulation removes the bitter taste and low bioavailability of extract. This study aims to determine the characteristics of ODF noni fruit extract and the immunomodulatory effects of ODF noni fruit extract in Wistar male rats induced by E. coli on the number of leukocytes, lymphocytes, monocytes, neutrophils, CD4 T cells, macroscopic and histopathological spleen. ODF evaluation included organoleptic, film thickness, film weight, folding endurance, percent elongation, disintegration time, and content uniformity. The rat immunomodulator test divided into normal grup, negative grup, positive grup, group 1 (extract 17.5 mg/KgBW), and group 2 (ODF extract). Treatment for 14 days, induction of E. coli 105 CFU/mL intraperitoneal on day 15 except the normal group. The evaluation results of ODF noni fruit extract had a thickness 0.128±0.001 mm, weight uniformity 0.082±0.001 mg, folding endurance 466.67±0.471, pH 6.10±0.014, percent elongation 33.042±0.010, disintegration time 33.667±0.471 seconds, and content uniformity 100.68±2.760%. ODF significantly decreased the number of leukocytes (34.88%), lymphocytes (34.72%), monocytes (52.48%), neutrophils (71.22%), and CD4 T cells (77.77%) than extract (p<0.05). ODF showed normal spleen, no necrosis, and no bleeding like in the positive group. Conclusion, ODF noni fruit extract has immunomodulatory activity as an immunosuppressant more effective than the extract.
