Abstract

Kencana Ungu (Ruellia tuberosa L.) leaves are known to contain secondary metabolites of flavonoids which have antidiabetic activity. The purpose of this research was to determine the effectiveness of reducing blood glucose levels of the ethyl acetate fraction of kencana ungu leaves. This study was divided into 6 groups, namely the normal group (0.5% Na-CMC suspension), positive control (1 IU/kgBB insulin), negative control (0.5% Na-CMC suspension) and treatment of the ethyl acetate fraction of kencana ungu leaves. with variations in doses of 50, 100, and 200 mg/kg BW. Alloxan induced rats with a dose of 125 mg/kg BW until blood glucose ≥ 126 mg/dL, ethyl acetate fraction of kencana ungu leaves was given for 14 days. The results showed that the percentage reduction in blood glucose levels of the ethyl acetate fraction of kencana ungu leaves at doses of 50, 100, and 200 mg/KgBW was significantly different from the negative control (p<0.05), with a percentage of 45.83% respectively; 49.78%; and 53.88%. The dose of 200 mg/kgBW did not differ significantly from insulin (p>0.05) while the doses of 50 and 100 mg/kgBW differed significantly from insulin (p<0.05). The ED50 of the ethyl acetate fraction kencana ungu leaves is 119.84 mg/KgBW. Pancreatic histopathological observations showed that the pancreatic β-cell necrosis score of rats at the 200 mg/KgBB dose had the best cell regeneration effect with a damage rate of 1-25% compared to the 50 and 100 mg/KgBB dose groups.
