Abstract

Peronema canescens (sungkai leaf) contains flavonoid secondary metabolites that show activity as immunostimulants. This study aims to determine the properties of Orally Dissolving Film (ODF) of sungkai leaf extract, immunomodulatory effects, macroscopic and histopathology of rat spleen. ODF was prepared using solvent casting method. Immunity parameters observed were CD4+ T cell values using PIMA analyzer and leukocytes, lymphocytes, neutrophils and monocytes using haematology analyzer. Histopathologic observation of the spleen using light microscopy with HE stain. The test animals used were male Wistar white rats with an average weight of 200 grams. The treatment was divided into 5 groups, namely normal (Feed), negative control (Placebo), positive control (Immunos), group I (0.4 mg/KgBB Extract) and group II (0.4 mg/KgBB ODF Extract). The treatment was carried out once a day for 7 days. Based on the results of the study, the evaluation results of the ODF extract properties were qualified. The results of mouse testing showed a significant increase in leukocytes in the extract and ODF extract groups (p<0.05). The results of the CD4+ value of the ODF extract group showed no significant difference with the positive control. Macroscopic observation of the spleen of the ODF extract group showed a normal spleen. The results of histopathology of the spleen of rats showed a mild degree of necrosis and hemorrhage. Based on the data of this study, it shows that Orally Dissolving Film (ODF) of sungkai leaf extract shows activity as an immunostimulant
