Abstract
Excision wounds have the potential to become infected when left untreated. Gadung stem (Dioscorea hispida) contains metabolites such as alkaloids, dioscorins and sapogenin that can accelerate the wound healing process. This study aims to determine the effect of ethanol extract of gadung stem wound healing activity rate. Twenty five Sprague Dawley rats were used as experimental animal which were divided into 5 groups; control negative groups, control positive group, and test groups K1, K2 and K3. Parameters observed included characterization extract of gadung stem, percentage of wound healing, duration of wound healing, and best dose ethanol extract of gadung stem to the wound healing process. Wound area data were analyzed using an ANOVA test to see the difference on a given day of observation. Phytochemical screening results showed ethanol extract of gadung stem (Dioscorea hispida) containing alkaloid compounds, flavonoids, saponins, steroid, and phenolics. The characterization ethanol extract of gadung stem (Dioscorea hispida) fulfills the requirements of the Indonesian Ministry of Health (2008). Determination of drying shrinkage, levels of soluble ethanol extracts, level of soluble water extracts, and level of total alkaloid (Dioscorea hispida) each is 6.48%, 88.9%, 77.05%, and 0.439%. The results showed that the extract of gadung stem with doses of 800 mg/kgBW had proven to accelerate wound healing as indicated by wound healing 100% arcived in rats in 20 days, show that the extract of gadung stem with doses of 800mg/kgBW had a significant difference to the control negative and positif group (Tekasol®). Then, there was a significant difference (p <0.05) between control negative group and treatment group (K2 and K3) on days 20th, it showed that positive control and treatment with dose variation can accelerate wound healing.
