Abstract

The river is a source of fresh water that serves to meet the vital needs of every living thing. River pollution is a problem in various regions, including in South Sumatra. The Komering River which passes through Ogan Komering Ilir district has a length of 80 km. Most people in the Kayuagung area still use the river to fulfill their daily needs. The location of the river is close to community activities such as sand mining, fish farming in cage systems, car washes, and other activities that will cause environmental problems and cause various kinds of infectious diseases caused by pathogenic bacteria. The use of antimicrobial agents often leads to resistance in microorganisms. The development of bacterial resistance to antibiotics needs to be monitored so that the treatment of disease with antibiotics can be done by selecting and dosing antibiotics correctly so that they are effective in curing the disease. Therefore it is important to do research, especially on Escherichia coli bacteria isolated from water bodies, especially the Komering River in the Rt 1 area of Kayuagung Asli Village, Kayuagung City. This study aims to determine the quality of Komering river water as seen from the density of Escherichia coli bacteria and to determine the resistance of Escherichia coli bacteria isolated from Komering River water to various types of antibiotics. This research was conducted from December 2022 to March 2023 at the Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The stages of the research consisted of prediction test, coliform confirmation test, E. coli prediction test, E. coli confirmation test, microscopic observation of E. coli bacteria under a microscope, IMVIC test, resistance test of E. coli bacteria to the antibiotics steptomycin, amoxicillin, chloramphenicol, cefixime, ciprofloxacin, sulfamethoxazole, ampicillin, tetracycline and kanamycin. The results showed that the frequency of the number of positive tubes in the coliform prediction and confirmation tests tested in Komering River water, Kayuagung Asli City, had a coliform count of >1,100 MPN/100 ml. The result is positive if gas is formed in the Durham tube. Meanwhile, the density of E. coli bacteria in Komering River water, Kayuagung Asli Village, was 2.9 MPN/ml. Based on IMViC testing, bacteria that form a metallic luster on EMBA medium are Escherichia coli bacteria. Characteristics of Escherichia coli bacteria microscopically observed under a microscope from the gram stain, namely rod-shaped, red in color, including gram-negative bacteria. Escherichia coli bacteria showed resistance to streptomycin antibiotics at concentrations starting from 0.01 µg /ml, amoxicillin at concentrations from 5-250 µg /ml, chloramphenicol starting from 0.01 µg /ml, cefixime at concentrations starting from 15 µg /ml, ciprofloxacin from 15 μg /ml concentration, sulfamethoxazole from 50 μg /ml, ampicillin from 5-250 μg /ml concentration, tetracycline from 10 μg /ml concentration, and kanamycin from 15 μg /ml. Determination of the nature of bacterial resistance is based on the inhibition zone formed around the antibiotic disc.
