Abstract

Azithromycin is a macrolide-derived antibiotic compound that is poorly water soluble. Various technologies are reported to overcome the solubility problem of poorly soluble drugs, one of which is solid dispersion technology. Solid dispersion refers to a group of solid products consisting of at least two different components, generally a hydrophilic matrix and a hydrophobic drug. Solid dispersions have the advantage of dispersing the drug in the carrier and converting the drug from a crystalline to an amorphous form to increase solubility, reduce particle size, and improve wettability. Chitosan can serve as a drug release enhancer for poorly water-soluble drugs due to the increased wettability resulting from the solubility of chitosan in water. The combination of the polymer with sodium alginate can increase the effectiveness of chitosan without producing systemic toxicity on its administration. This study aims to determine the effect of chitosan and sodium alginate as polymers on the dissolution rate of azithromycin solid dispersion. The co-grinding method was chosen as a technique for the preparation of azithromycin solid dispersion due to its easy handling, affordable cost, and environmental friendly. Three formulations of azithromycin solid dispersion were made with varying amounts of sodium alginate. Evaluation and characterization of azithromycin solid dispersion samples were conducted through XRD, FTIR, SEM, solubility test, and dissolution rate test. The test results showed a decrease in the intensity of azithromycin crystals, the absence of chemical interactions between azithromycin and polymers, changes in solid dispersion morphology, solubility of solid dispersion in SIF solution, and an increase in the dissolution rate of azithromycin in solid dispersion. Based on the test results, it can be concluded that the preparation of azithromycin solid dispersion by co-grinding method can increase the solubility and dissolution rate of azithromycin.
