Abstract
Waste is a pollutant that can damage the ecosystem. One of the contents of the waste is the heavy metal cadmium (Cd). The existence of cadmium waste can be dangerous so it is necessary to control environmental pollution. An alternative solution in efforts to control cadmium waste pollution is to carry out bioremediation using endophytic bacteria isolated from the roots of the water hyacinth plant (Eichhornia crassipes (Mart) Solms.). Resistance mechanisms possessed by endophytic bacteria are in the form of biosorption and bioaccumulation which will help the process of restoring polluted environments by reducing heavy metal toxicity. The purpose of this study was to determine endophytic bacteria that are potentially resistant to cadmium so that they can be used as bioremediation agents for environments polluted by cadmium waste. This research was conducted from December 2022 to March 2023 at the Microbiology Laboratory, Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. The stages of this research consisted of isolating endophytic bacteria from water hyacinth roots, purifying endophytic bacteria, macroscopic observations, microscopic observations with gram and endospore staining, resistance testing using the pour plate method and calculating the number of bacterial colonies using the TPC method in media containing cadmium with a concentration of 30, 40, 50, and 60 ppm, as well as characterization and identification of endophytic bacterial isolates that are resistant to cadmium. The results of this study were 4 isolates of water hyacinth root endophytic bacteria that were resistant to cadmium. Isolate EG. 2 had the best resistance to cadmium with a colony count of 7.8 x 109 CFU/mL at a concentration of 60 ppm. Isolate EG. 7 had good resistance to cadmium with a colony count of 4.6 x 109 CFU/mL at a concentration of 60 ppm. Isolate EG. 9 and EG. 4 had poor resistance to cadmium with the number of colonies respectively 9.0 x 106 CFU/mL and 3.4 x 106 CFU/mL. Isolate EG. 2 was suspected from the genus Corynebacterium, isolate EG. 4 was suspected from the genus Staphylococcus, while isolate EG. 7 and EG. 9 were suspected from the genus Neisseria.
