Abstract

Ibu menyusui yang terpapar asap rokok dapat terkontaminasi nikotin, terutama pada ASI yang dihasilkan oleh Ibu menyusui. Penelitian ini bertujuan untuk memvalidasi metode analisis kadar nikotin dalam ASI dengan metode GLC serta menentukan dan membandingkan kadar nikotin dalam ASI ibu perokok pasif yang sedang menyusui berdasarkan lamanya terpapar (1-15 tahun). Validasi metode dilakukan dengan menentukan linearitas, LOD, LOQ, akurasi, presisi dan ketidakpastian pengukuran hasil analisis kadar nikotin dalam ASI yang mana diperoleh persamaan regresi linear yaitu y = 289166x - 383,48 dengan r = 0,9979, LoD sebesar 0,003976 ppm dan LoQ sebesar 0,013255 ppm. Adapun persen perolehan kembali pada sampel dengan penambahan larutan standar 0,01 ppm sebesar 31,92%, kemudian persen RSD sebesar 0,32-4,27% dengan estimasi ketidakpastian pengukuran sampel 1 0,014 ± 0,0026522 ppm; sampel 2 0,012 ± 0,0026519 ppm; sampel 3 0,016 ± 0,0026526 ppm; sampel 4 0,015 ± 0,0026524 ppm; sampel 5 0,017 ± 0,0026529 ppm. Hal ini menunjukkan bahwa dari beberapa parameter validasi metode analisis, hanya persen perolehan kembali yang tidak memenuhi syarat keberterimaan atau dengan kata lain metode tidak valid apabila didasarkan pada parameter uji akurasi. Kadar nikotin yang terukur pada sampel ASI ibu perokok pasif dengan range lama terpapar 3-13 tahun berkisar dari 3,71-11,59 ug/L dengan nilai terendah 3,71 ug/L yang dimiliki oleh sampel 2 dengan lama terpapar 4 tahun, sedangkan nilai tertinggi sebesar 11,59 ug/L yang dimiliki oleh sampel 5 dengan lama terpapar 3 tahun. Hal ini menunjukkan bahwa semakin lama terpapar asap rokok, belum tentu jumlah nikotin di dalam ASI akan semakin banyak.
