Abstract
Research on yeast isolation and identificatin of durian fruit has been conducted for fermentation the glucose and starch to produce ethanol. The yeast isolates D1, D2, D3, D4, D5, and D6 obtained from 10-2 dilutions were grown on YPDA (Yeast Peptone Dextrose Agar), YPAA (Yeast Peptone Arabinose Agar), YPXA (Yeast Peptone Xylose Agar) and YPSA (Yeast Peptone Cellulose Agar). The growing yeast isolates are further analyzed to be identified microscopically, macroscopically, molecullary and with the API Kit 20C AUX method. Based on observations using a microscope showing that isolates D1, D2, D3, D4, and D6 have a round cell shape, isolate D5 has a bacillus form. The results of identification using API Kit 20C AUX showed that the isolates obtained were Candida guilliermondii, Candida boindinii, Candida albicans, Cryptococcus terrus, Pichia Angsta, and Cryptococcus uniguttulatus. Isolates that have been identified are used for the fermentation of glucose and hydrolyzed substrate using 10% H2SO4. Based on gas chromatography measurement data, it was found that the highest ethanol content in glucose substrate was found in fermentation using isolate Candida guilliermondii on the third day of 0.0000334%. For the fermentation of the highest starch ethanol content was found in isolate Candida guilliermondii on the first day of 0,0000231%.
