Abstract

Oil palm empty fruit bunches (OPEFB) is a by-product with high potential as a source of cellulose. This study aims to 1) screen cellulase-producing bacteria from OPEFB waste using selective media Carboxy Methyl Cellulose (CMC) 1% clarified with congo red 0.1% to measure cellulolytic index values, 2) test cellulase enzyme activity using a sugar test reduction of the Dinitro salicylic acid (DNS) method and 3) compare the results of the 2 methods. Based on the clear zone, 20 isolates of cellulolytic bacteria were found, the isolates were retested using the reducing sugar method to determine the value of their enzyme activity using DNS. The data obtained is presented quantitatively and descriptively. The seven isolates that had the highest activity values were further tested, including the morphology of the bacterial isolates, the catalase test, the motility test, the H2S test and Gram staining. The seven isolates that had the highest cellulolytic index values was in isolates D 10-6(47), A 10-6(79), C 10-7(39), A 10-7(20), D (10-6)54, D 10-6(51) and D 10-6(48) namely of 8.92, 5.27, 4.02, 3.35 ,3.29, 2.97 and 2.71. While the best isolates based on the reducing sugar test was in isolates C 10-7 (39), D 10-6 (44), D 10-6(56), D 10-6(79), D 10-6(46), D 10-6(54) and D 10-6(50) with enzyme activity values of 0.901, 0.871, 0.736, 0.719, 0.718, 0.671 and 0.596 (U/mL). Differences in the results of the clear zone bacterial screening method and the DNS method enzyme activity test can be affected by temperature and incubation time during enzyme production and the use of non-refrigerated centrifuges. Based on Gram staining, all isolates had a bacil cell shape and were classified as Gram-positive bacteria, except for A 10-6 (79) which were classified as Gram-negative bacteria. All isolates positively produced catalase, motile, capable of producing reducing sugars and negatively produced H2S and CO2.
