Abstract
Lansium domesticum Corr. or other names for duku and langsat are germplasm plants that have high economic value and have the potential, especially in South Sumatra, to become the mascot of flora in South Sumatra. In order not to lose these species, it is necessary to carry out a conservation effort with the aim of knowing the genetic variation and kinship of Lansium domesticum Corr. in South Sumatra. This research was conducted using 4 primers from Random Amplified Polymorphic DNA (RAPD) markers, namely primers OPA 02, OPB 07, OPU 19 and OPU 20 with 11 samples of Lansium and 1 sample of Aglaia from South Sumatra. The four primers were able to amplify as many as 83 DNA bands with a high level of polymorphism of 98.8% and also showed high genetic variation. The results of the genetic similarity matrix show that the closest kinship is in the accessions of Duku Pali and Duku Lahat, while the furthest kinship is in the accessions of Duku Baturaja and Langsat Linggau. The results of the dendogram reconstruction obtained two main clusters, namely the duku cluster and the langsat cluster with a coefficient value of 0.22 - 0.83 with the Jaccard Coefficient (Sȷ) and 0.39 - 0.92 with the Simple Matching Coefficient (Sѕм). Lansium domesticum Corr. atau nama lain dari duku dan langsat merupakan tanaman plasma nutfah yang memiliki nilai ekonomi tinggi dan berpotensi khususnya di Sumatera Selatan sehingga menjadi maskot flora di Sumatera Selatan. Agar tidak hilangnya spesies tersebut maka perlu dilakukannya suatu upaya konservasi dengan tujuan untuk mengetahui variasi genetik dan hubungan kekerabatan Lansium domesticum Corr. yang ada di Sumatera Selatan. Penelitian ini dilakukan dengan menggunakan 4 primer dari marka Random Amplified Polymorphic DNA (RAPD) yaitu primer OPA 02, OPB 07, OPU 19 dan OPU 20 dengan 11 sampel Lansium dan 1 sampel Aglaia dari Sumatera Selatan. Keempat primer mampu mengamplifikasi sebanyak 83 pita DNA dengan tingkat polimorfisme yang tinggi sebesar 98,8% dan menunjukan variasi genetik yang tinggi pula. Hasil matriks similaritas genetik menunjukan hubungan kekerabatan terdekat pada aksesi Duku Pali dan Duku Lahat sedangkan hubungan kekerabatan terjauh pada aksesi Duku Baturaja dan Langsat Linggau. Hasil dari rekontruksi dendogram didapatkan dua klaster utama yaitu klaster duku dan klaster langsat dengan nilai koefisien sebesar 0,22 - 0,83 dengan Jaccard Coefficient (Sȷ) dan 0,39 - 0,92 dengan Simple Matching Coefficient (Sѕм).
