Abstract

SALMAN KHAERUL HAKIM, Antibacterial Activity Of Watercress (Nasturtium Officinale) Ethanol Extract Against Staphylococcus Aureus and Pseudomonas Aeruginosa Bacteria (Supervised by SABRI SUDIRMAN). Bacteria are microorganisms that can cause quality degradation in fishery products and cause disease in humans. Borax and formalin are commonly used antibacterial agents, but they have a negative impact on health. Polyphenols from plant extracts are a source of natural antibacterials. This study aimed to determine the effectiveness of watercress (Nasturtium officinale) ethanol extract against Staphylococcus aureus and Pseudomonas aeruginosa bacteria. The polyphenolic compounds were extracted by the maceration method using 70% ethanol; the level of polyphenolic content was determined by the Folin–Ciocalteu method; and the antibacterial activity was determined by the Kirby-Bauer disc diffusion method, each of which was carried out 3 times. The yield of the extract obtained was 21.62 ± 1.55% with a polyphenol content of 28.49 mg GAE/g dry sample. Antibacterial activity shows that an increase in sample concentration directly proportional to the inhibition of each bacterium. The diameter of the inhibition zone at a concentration of 1 mg/mL for Pseudomonas aeruginosa was 11.13 mm and for Staphylococcus aureus was 9.05 mm. Based on these results, the polyphenolic compounds from watercress can be used as natural antibacterial agents. (Dibimbing oleh SABRI SUDIRMAN). Bakteri merupakan mikroorganisme yang dapat menyebabkan pembusukan pada produk perikanan dan menyebabkan penyakit terhadap manusia. Boraks dan formalin merupakan agen antibakteri yang umum digunakan, namun memiliki dampak negatif bagi kesehatan. Polifenol dari ekstrak tumbuhan merupakan salah sumber antibakteri alami. Penelitian ini bertujuan menentukan efektivitas antibakteri ekstrak etanol selada air (Nasturtium officinale) terhadap bakteri Staphylococcus aureus dan Pseudomonas aeruginosa. Senyawa polifenol diekstrak dengan metode maserasi menggunakan etanol 70%, kadar polifenol ekstrak ditentukan dengan metode Folin–Ciocalteu, dan aktivitas antibakteri dengan metode difusi cakram Kirby-Bauer yang masing-masing dilakukan pengulangan sebanyak 3 kali. Rendemen ekstrak diperoleh sebesar 21,62±1,55% dengan kadar polifenol sebesar 28,49 mg GAE/g sampel kering. Antibakteri menghasilkan peningkatan konsentrasi sampel berbanding lurus penghambatan masing-masing bakteri. Diameter zona hambat pada konsentrasi 1 mg/mL pada Pseudomonas aeruginosa yaitu 11,13 mm dan Staphylococcus aureus yaitu 9,05 mm. Berdasarkan hasil tersebut, senyawa polifenol dari selada air dapat dijadikan sebagai agen antibakteri alami.
