Abstract
Buffalo milk dadih is a traditional fermented milk from West Sumatra. In the fermentation process, Dadih contains Lactic Acid Bacteria (LAB), which produce secondary metabolites such as organic acids (lactic, acetic, and formic acids), hydrogen peroxide, diacetyl, acetaldehyde, acetoin, reutinin, reutericyclin and bacteriocin. The resulting compound can act as an anti-Candida agent that protects the body by preventing the growth of pathogenic microbes that cause infection. C. parapsilosis is widely used as a test fungus in antifungal agents to prevent infection in the human body. Further research on the ability of LAB in buffalo milk curd to produce antifungal compounds needs to be carried out to find out how much the ability of the antifungal compounds produced by LAB isolates from buffalo milk curd can inhibit the growth of Candida parapsilosis. This research was conducted from January 2023 to March 2023 at the Microbiology Laboratory, Physiology and Development Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The stages of the research consisted of isolation of LAB from buffalo milk curd, purification of LAB isolate, rejuvenation of the tested fungus (C.parapsilosis), cultivation of LAB, preparation of a suspension of the tested fungus (C.parapsilosis), testing of the antifungal activity of LAB isolates using the paper disk method and dual agar overlay, inhibition zone measurement and characterization. Based on the results of the antifungal activity of LAB isolates from buffalo milk curd using the dual agar overlay method, it was shown that all isolates could inhibit C. parapsilosis as indicated by the formation of clear zones that varied in each LAB isolate. The clear zone variation indicated that each LAB isolate could produce different antifungal compounds. Meanwhile, based on the results of the antifungal activity of LAB isolates from buffalo milk curd using the disc diffusion method, it was shown that three isolates had the greatest ability to inhibit the growth of C.parapsilosis, namely LAB 7 isolates, LAB 12 isolates and LAB 14 isolates with the resulting inhibition zone diameter values respectively (4.83 mm ± 0.28), (4.33 mm ± 0.28) and (4.66 mm ± 0.57). Based on the results of the characterization and biochemical tests, it was found that the three identified LAB isolates belonged to the genus Lactobacillus.
