Abstract

The bintaro plant (Cerbera manghas L.) is a poisonous plant of the Apocynaceae family that has potential as an anticancer agent because it contains secondary metabolite compounds that show cytotoxic activity which can be known based on the level of toxicity. This study aims to determine the acute toxicity of n-hexane extract, ethyl acetate extract, and methanol extract of bintaro fruit on the mortality of Artemia salina Leach larvae using the brine shrimp lethality test (BSLT) method and also to determine the secondary metabolite compounds contained in bintaro fruit methanol extract using liquid chromatography-mass spectrometry (LC-MS) method. This research was conducted in August 2022-December 2022 and sampling was carried out in Bogor Regency, West Java Province. Acute toxicity test of bintaro fruit extract was conducted at the Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. LC-MS analysis was conducted at the Bogor Police Forensic Laboratory Center. The stages of this research consisted of preparation and making of simplisia, multistage extraction process, making artificial seawater, hatching Artemia salina Leach eggs, acute toxicity test of bintaro fruit extract, acute toxicity analysis, LC-MS analysis and data analysis. This acute toxicity test study was divided into 5 negative control groups and 5 concentration variations of n-hexane, ethyl acetate and methanol extracts. Each concentration consisted of 10 larvae with 3 replications for each treatment. The five treatment groups were given bintaro fruit extract with concentrations of 62.5 ppm, 125 ppm, 250 ppm, 500 ppm, and 1,000 ppm. The results showed the LC50 value of n-hexane extract of bintaro fruit was 81.34316 µg/ml, ethyl acetate extract was 101.9093 µg/ml and methanol extract was 71.34806 µg/ml. Based on this acute toxicity test, it is known that the three types of extracts have cytotoxic activity and can act as anticancer agents because they have LC50 values <1,000 µg/ml. Based on LC-MS analysis conducted on methanol extract of bintaro fruit, it proves that bintaro fruit contains secondary metabolite compounds in the form of cerberin and neriifolin from ten known metabolite compounds and has cytotoxic activity.
