Abstract

Background: Acute gouty arthritis is an inflammation caused by MSU crystal deposition in the joint. Kersen leaves contain flavonoids that have anti-inflammatory and antioxidant activities. This study aims to determine the effect of kersen (Muntingia calabura L.) leaf extract on the histopathological profile of gout model rat joints. Methods: This research is an in vivo post-test-only control group experimental study. Kersen leaf ethanol extract (EEMC) was prepared by macerating kersen leaf simplicia with 96% ethanol. Thirty male Wistar rats were divided into five groups: normal control, negative control (Na-CMC 0.5%), and three groups of kersen leaf extract doses of 200, 400, and 800 mg/kgBW. Rats in the negative control and extract groups were injected by MSU crystal suspension intraarticularly to make a gout model. Rats were terminated on the third day, and knee joints were taken for HE slides. A histopathological examination was performed by assessing the synovial inflammation and cartilage erosion profile scores. The data were then analyzed using SPSS with a statistical significance of p < 0,05. Results: The synovial inflammation and cartilage erosion profile scores obtained from each group, respectively, were 0,67 and 0,67; 3,17 and 6,00; 3,33 and 2,17; 1,67 and 0,83; and 1,83 and 1,33. Between groups, there were significant differences in cartilage erosion profile scores and non-significant differences in synovial inflammation profile scores. Conclusion: EEMC has an effect on improving the cartilage erosion profile score but has no effect on improving the synovial inflammation profile score in the knee joint of gouty rat models.
