Abstract

Spiny eel (Mastacembelus sp.) is a freshwater fish that is distributed on the islands of Java, Sumatera and Kalimantan. Mastacembelus erythrotaenia has been found in the Musi River and Mastacembelus unicolor has been found in the Enim River, the two species have different colors and patterns on their bodies. Therefore, it is necessary to study the genetic characteristics of these two species. This research aims to obtain the quantity and quality of spiny eel DNA, determine the COI gene sequence of spiny eel mitochondrial DNA and the percentage of similarities between spiny eel and other fish in GenBank data, as well as determine the water quality in the spiny eel habitat. This research was carried out in February to August 2023. The methods used in barcode were DNA extraction, DNA amplification, electrophoresis and sequencing of the COI gene region. The PCR product was obtained from the amplification with an annealing temperature of 49.2°C for 45 seconds in 31 cycles. The results showed that the DNA quantity value of spiny eel from the Musi and Enim Rivers ranged from 55.1-131.9 ng μL-1, the absorption ratio of A260/A280 from 1.64-1.98 and A260/230 from 1.60-2.73. The sequence of PCR products produced nucleotide lengths from 207-388 bp. The percentage of similarities between spiny eel and other fish in GenBank data cannot be determined. Transparency values ranged from 21.25-42 cm, dissolved oxygen (DO) from 3.52-7.66 mg L-1, current velocity from 0.14-0.47 m s-1, alkalinity from 20-40 mg L-1, pH from 6.57-7.6, temperature from 25.2-28.9°C, total dissolved solid (TDS) from 11-15 mg L-1, ammonia from 0.20-0.59 mg L-1.
