Abstract

Background : Indonesia was ranked 5th worldwide regarding diabetic patients with a total number of 19,5 millions, and is expected to increase by around 21.3 millions by 2045. 90% of the case is a Type 2 Diabetes Mellitus that were caused by insulin resistance, which can be triggered by IL-6. Karamunting (Rhodomyrtus tomentosa) is a plant that often be used as a herbal medicine. Quercetin is one of many compounds found in Karamunting with medical properties such as anti-inflammatory and antioxidant. The advancements of the bioinformatic field facilitates the drug development field also. This study aims to determine how bioinformatically the potential target of quercetin compounds from the Karamunting plant (Rhodomyrtus tomentosa) and its interaction with the cytokine IL-6 which plays a role in one of the pathogenesis of insulin resistance. Methods : The research method used in this research is descriptive explorative with pre-experimental in silico design. Result : Based on the prediction result of Lipinski's rule of five, quercetin met all the criteria of druglikeness. The docking result shown that the binding affinity of quercetin and Tnf-a complex was -7.1 kcal/mol, which is lower than the native ligand that is around -3.8 kcal/mol. It is shown by the docking visualization that quercetin had a little resemblances in the amino acid residues interaction compared to that of the native ligand. Conclusion : The molecular docking result has shown that quercetin does have a lower binding affinity than the native ligand, but the interaction results were not so similar. Nevertheless, quercetin still had the potential to be developed as a IL-6 blocker. Quercetin also passed the druglikeness test so that it has the potential to be an oral medicine
