Abstract
Ceratocystis has been reported disease-causing pathogen in Acacia mangium, which is from the legume family. Endophytes are defined as microorganisms that live and develop in plant tissue organs and are able to increase plant resistance to disease and environmental stress. In 2022, biological control has been carried out by exploring endophytic and non-endophytic fungi from the stems and roots of duku plants by Fadillah 2022; Pratiwi 2022; Triagtin 2022, but the research has not identified species that have the potential to suppress the pathogen Ceratocystis fimbriata and has not made microscopic observations of interactions that occur between pathogenic and antagonistic microbes. The purpose of this study was to further determine the species of endophytic fungi in duku plants in South Sumatra and their potential to suppress the growth of Ceratocystis pathogen that causes wilt disease of duku plants. The benefits of this study are to determine the species of endophytic fungi present on the stems of duku plants and their potential in suppressing the growth of the pathogen Ceratocystis which causes wilt disease in duku plants. This research was conducted at the Phytopathology Laboratory, Faculty of Agriculture, Sriwijaya University. The research began in June 2023 until completion. Endophytic fungal isolates used are the result of exploration in previous studies. These isolates came from the stems of duku plants in Ogan Ilir Regency, Ogan Komering Ilir Regency, and Musi Banyuasin Regency. This study used a completely randomized design (CRD) with 3 replicates. The results of this study showed that endophytic fungal isolates from the stem of the duku plant that had been isolated were fungi from different genus. Endophytic fungal isolates that have been identified come from the genus Penicillium sp, Aspergillus sp, Fusarium sp, Aureobasidium sp, Phoma sp, Madurella spp, and several isolates (SDW 5A, DSPTW 9A, TRW 5, and WPN 4B whose genus is unidentified. 11 isolates of endophytic fungi showed different inhibitory power, isolates with the highest inhibitory power were SDW 3B (52%), isolates with moderate inhibitory power were isolates of DSPTW 6A, DSPTW 7B, DSPTW 8A, and SDW 5A (44% to 47%) and the lowest was isolate DSPTW 9A (39%). The mechanism of hyphal interaction was found in 6 isolates (SDMW 7B, SDW 3B, RMIP 1, TRW 5, TRW 1B and WSA 4A) which caused pathogenic hyphae to lysis. 2 isolates (DSPTW 6A and DSPTW 9A) that cause pathogenic hyphae to swell. 4 isolates (SDW 5A, SDMW 8A, TRW 1B, and WPN 4B) caused pathogenic hyphae to twine and lysis.
