Abstract

Rubber plants (Hevea brasiliensis Muell.) are one of Indonesia's plantation commodities. Rubber plays an important role as a source of income for farmers in Indonesia and is mostly used as a source of making rubber materials (latex), industrial materials and making asphalt. The low production of rubber plants can be influenced by factors from plant disrupting organisms, one of which is a disease from the fungal group. One of the causes of these losses is rubber leaf fall disease caused by Pestalitopsis sp. This study aims to test the ability of endophytic bacteria derived from rubber plants to inhibit the growth of leaf fall disease Pestalotiopsis sp. and endophytic bacteria have the potential as a safe biofungicide. This study consists of two series. The first series involves testing the antagonistic effects of endophytic bacteria (from the Central Research Laboratory of Sembawa Rubber Research Center) using a Completely Randomized Design (CRD) with 23 treatments of endophytic bacterial isolates from rubber plants and one control, each with 5 replications, resulting in a total of 120 Petri dishes. The second series includes hypersensitivity tests conducted on 23 endophytic bacterial treatments and hemolysis tests performed on 7 endophytic bacteria. The results obtained 23 isolates of endophytic bacteria from rubber plants that have varied colony morphological characteristics and biochemical tests that show diversity. Endophytic bacteria have the potential as biofungicides to control leaf fall disease Pestalotiopsis sp. Antagonistic testing showed the ability of endophytic bacteria to inhibit the growth of the pathogenic fungus Pestalotiopsis sp. Isolate PNB32 stood out with the highest inhibition on the sixth and seventh days reaching 54.94 percent. Microscopic observation of abnormal growth in fungal hyphae, endophytic bacteria caused abnormal growth in hyphae, namely stunted, swollen, shortened and truncated hyphae. Hypersensitive reactions in tobacco plants there are 10 bacteria that are pathogenic including B2, B5, B19, B37, B40, B42, B45, B52, B54, and B56. Biomass weight of Pestalotiopsis sp. the lowest value in isolate PNB21 which is significantly different from PNB4 and PNB1. This decrease in biomass can be attributed to the growth of thin mycelium on NA media, resulting in low biomass weight of Pestalotiopsis sp. fungi. The results of hemolysis testing on Blood Agar showed that only one endophytic bacterial isolate did not degrade blood agar cells, so it has potential as a safe biofungicide.
