Abstract

Erythromycin belongs to Biopharmaceutical Classification System (BCS) class 2, indicating limited solubility, which in turn results in restricted penetration and effectiveness.Incorporating erythromycin into a nanoemulsion delivery system can be a promising approach to overcome its limitations. This study aims to find the optimum formula for erythromycin nanoemulsion with variations in the concentration of the active substance and Virgin Coconut Oil (VCO) as the oil phase, using Design Expert software with a factorial design method 22 . Erythromycin nanoemulsion was created using a combination of high-energy process methods. The optimal formula was determined based on the highest desirability value with the best characterization results, including %EE (%Encapsulation Efficiency), %transmitance, viscosity, pH, density, and stability tests. System optimization results indicated that the optimum concentrations of erythromycin and VCO were 2% and 5%, respectively. Characterization of the optimum formula of erythromycin nanoemulsion resulted in a particle Size of 143.833 ± 3.083 nm, a polydispersity index value of 0.368 ± 0.00, and a zeta potential of -16.05 mV ± 0.00. The optimum formula of erythromycin nanoemulsion also demonstrated good physical stability based on cycling and centrifugation test results, with no phase separation and significant pH changes. Based on the results obtained, the optimum formula of erythromycin nanoemulsion has good characterization and stability results in accordance with the literature.
