Abstract

Erythromycin is a drug classified into Biopharmaceutical Classification System (BCS) class II which has low solubility in water so that it can reduce the speed of drug penetration into skin. One of the efforts to increase the penetration of Erythromycin is to formulate erythromycin into a nanoemulsion delivery system. The nanoemulsion was prepared by the PIT (Phase Transition Temperature) method which is a development of the spontaneous emulsification and low energy stirring methods, then using ultraturrax to reduce droplet size and ultrasonication to maintain droplet size. This study aims to find the optimum formula of Erythromycin nanoemulsion with varying concentrations of Tween 80 and PEG 400 as surfactant and cosurfactant using Design Expert software with Factorial design 22 method. The optimum formula was determined based on the results of organoleptic test characterization, entrapment efficiency (%EE), percent transmittance, viscosity test, pH test, density, and stability test. The optimum formula selected based on the system is a nanoemulsion that has a concentration of tween 80 25% and PEG 400 25% which has a desirability value <1. The characterization results of the optimum formula obtained a particle size of 170,6 ± 12.8594 nm, a polydispersity index (PDI) of 0,403 ± 0,04406, and a zeta potential of -8,8667 ± 0,25697 mV and had good stability because it did not experience phase separation in the Centrifuge test and Cycling Test, and only decreased pH by 0,37% in each cycle. Based on the results obtained, the optimum formula of erythromycin nanoemulsion has good characterization and stability.
