Abstract

Mefenamic acid is a non-steroidal anti-inflammatory drug (NSAID) that belongs to biopharmaceutical classification system (BCS) class II. Mefenamic acid has a very poor solubility that requires formulation strategies that can increase the dissolution rate. Solid dispersion systems with hydrophilic polymer matrix carriers have been adopted as a formulation strategy to improve the solubility of active substances. This study aims to determine the effect of sodium alginate polymer amount on the characteristics and evaluation of mefenamic acid/sodium alginate/chitosan solid dispersion. The variation of sodium alginate amount used was 200 mg, 250 mg, and 300 mg. Mefenamic acid/sodium alginate/chitosan solid dispersion was prepared using co-grinding method. Characterization and evaluation included XRD, FTIR, SEM, and dissolution tests. The results of characterization and evaluation showed that the ideal amount of sodium alginate was 250 mg which was characterized by decreasing crystallinity degree, without any significant change in molecular structure, phase change from crystalline to amorphous, and increased dissolution rate. Solid dispersion of mefenamic acid/sodium alginate/chitosan prepared by co-grinding method increased the solubility of mefenamic acid.
