Abstract

Bitter leaf extract (Andrographis paniculate (Burm.f) Wall. Ex Ness) contains secondary metabolites andrographolide and quercetin which have potential antibacterial activity. This research aims to determine the nanoemulsion formulation with a combination of Tween 80 and Ethanol 96% concentration on the activity of Propionibacterium acnes and the stability of ICH Q1A (R2). Data analysis in this study used the One Way ANOVA method in the SPSS for Windows program by observing significant differences in the response of the test parameters. The bitter nanoemulsion uses Tween 80 and Ethanol 96% with the same concentration, namely 25%. The resulting bitter nanoemulsion has organoleptic characteristics, a light green color and a distinctive aroma of bitter and basil oil, pH of 6.533 ± 0.011, viscosity of 4.943 ± 0.020, specific gravity of 0.994 ± 0.001, percent transmittance of 99.333 ± 0.057, adsorption efficiency of 92.368 ± 0.057, andrographolide levels were 202.252 ± 0.100, quercetin levels were 18.474 ± 0.047, globule size was 104.67 ± 11.95, PDI was 0.462 ± 0.065, zeta potential was -7.866 ± 0.318. The stability results with the ICH Q1A (R2) standard state that the nanoemulsion formula has a first-order release model, activity energy of 20,434 cal/mol and has a shelf life at 25oC conditions for 84,2696 days and 5oC days for 142,4093 days. The nanoemulsion was also tested for antibacterial activity using the disc diffusion method and determining the MIC. The diameter value of the nanoemulsion zone was 12,366 ± 1,184 mm and 1% bitter extract was 8,666 ± 1,106 mm. Meanwhile, the MIC value of nanoemulsion was at a concentration of 80mg/ml and 1% bitter extract was at a concentration of 160mg/ml. Based on the research results, it was concluded that the nanoemulsion preparation had good characteristics and stability as well as strong antibacterial activity compared to bitter leaf extract which was not formulated in nanoemulsion.
