Abstract

Lansium domesticum or duku plant is a native Indonesian fruit plant that is widely cultivated in South Sumatra. Duku plants are rich in benefits from both health and agricultural aspects. In 2014 there was a sudden wilt attack of duku plants in the Ogan Komering Ulu (OKU) area and spread to several other districts such as Muara Enim. Observations on the causes of this disease in Muara Enim Regency have never been done before. This study aims to observe the cause of wilt disease in duku plants in Rami Pasai, Padang Bindu, Betung and Pagar Dewa villages, determine the characteristics of pathogen isolates that cause wilt disease of duku plants and determine the results of Koch's postulate test and host range test on 10 types of plants to be tested. This research was conducted at the Phytopathology Laboratory, Faculty of Agriculture, Sriwijaya University by exploring microorganisms carried out in 4 villages in Muara Enim Regency, South Sumatra, namely in the villages of Rami Pasai, Betung, Padang Bindu and Pagar Dewa. Sampling in this research used Purposive Sampling method in each village observed and then isolated pathogens from duku stems on MEA and V8 juice media. Furthermore, Koch's postulate test was carried out on healthy duku plants as well as host range and pathogenicity tests of 4 isolates of Ceratocystis sp. fungus on 10 types of test plants, namely duku, durian, cape, avocado, mahogany, kabau, jelutung, jengkol, petai and red shoots with the Complete Randomized Design (CRD) method in the shadow house. The observed variables included the growth of pathogen isolates on V8 juice media, morphology of fungi that cause wilt in duku plants, symptoms of pathogen attack in Koch's postulate test and host range and lesion length on test plants. Data analysis using ANOVA (analysis of variance) Excel 2019 and SPSSsoftware. The results obtained are the cause of wilt disease in duku plants in Muara Enim Regency is the fungus Ceratocystis sp. There are similarities in disease symptoms from each village, namely the shape of scratches and spheres, yellowing of leaves and wilting of the stem. Microscopic identification results show Ceratocystis has a morphology of globose peretecium bases with long necks, divergent ostiolar hyphae, conidiophores or phialide, chlamydospores, capshaped ascospores, filamenteus and moderate mycelial growth patterns with grayish black and brownish black colonies. The isolation results showed that the fungal growth was relatively slow (1,39-3,16 cm) within 5 days. Koch's postulate test showed that all isolates causing wilt disease on duku plants were not significantly different between treatments with the length of lesions formed, namely 8,63 ± 1,87 b to 9,31 ± 1,46 b with the number of dead test plants, namely 3-5 plants. The host range test shows that this disease can infect other plants with the same symptoms caused in the field.
