Abstract

Erythromycin is a topical antibiotic with anti-inflammatory and immunomodulatory activities used for the therapy of mild to moderate inflammatory acne. However, conventional topical erythromycin formulations are still ineffective as they struggle to penetrate deeper into the sebaceous areas located in the dermal layer of the skin. Therefore, it is necessary to develop a topical formulation incorporating nanotechnology features, such as the modification of nanoemulsion formlations. This research aims to optimize the nanoemulsion formula of erythromycin by varying the concentrations of Virgin Coconut Oil (VCO) and tween 80 using the factorial design method 2^2 through the Design-Expert 12® tool. The study employs four formulas, and the optimal formula is determined based on characterization results, including pH value, specific gravity, viscosity, percent transmittance, absorption efficiency, and stability tests. The stability test results using the cycling test method indicate that formula 2 with concentrations of VCO and Tween 80 at 5% and 25%, respectively, produces the best physical stability with a fairly stable pH value during storage. The selected optimal formula, based on the system, is at a concentration of 5% Virgin Coconut Oil (VCO) and 25% Tween 80. The characterization results of the optimal formula show a particle size of 143.367 ± 3.406, polydispersity index of 0.352 ± 0.013, and zeta potential of -10.32 ± 0.058. Based on the obtained results, the optimal nanoemulsion formula of erythromycin has shown good characterization results and fairly good physical stability.
