Abstract

Spirulina platensis is a blue-green algae species that contains phycocyanin. The low stability of phycocyanin requires an effective delivery system, one of which is through the process of nanoencapsulation using the ionic gelation method. Potential natural polymers serving as encapsulating agents include chitosan and alginate. Folic acid is among the targeting moieties used for the targeted delivery of phycocyanin. The study aimed to characterize the conjugation outcomes with folic acid ligands in each formulation, determine the concentrations of chitosan and alginate to produce the optimal formulation of chitosan-alginate nanoencapsulation carrying phycocyanin conjugated with folic acid, evaluate the stability and in vitro release of the optimal formula. This research commenced with the characterization of phycocyanin of λmax, content & purity, FT-IR and SDS-PAGE, the preparation of folic acid-conjugated chitosan (FA-C) and formulation (P-FA-C-Al-NP) along with their characterization, then determination of the optimal formulation and its evaluation. Data analysis using Design-Expert 12® employing the Factorial Design 22 method and SPSS software. The research showed that ficocyanin λmax was 616 nm with a content of 10.3 mg/g and a purity value of 2.8. SDS-PAGE testing shows the presebce of α and β polypeptide subunit with molecular weight of 14.4-18.4 kDa and 18.4-25.0 kDa. FT-IR results show that has absorption in the form of amide (N-H) functional groups at 3364.09 cm-1, carboxyl (C-O) groups at 1656.77 cm-1, and absorption of 1000-1500 cm-1 which indicates a pyrrole ring. The optimal concentrations of chitosan and alginate in the formulation were determined to be 0.3% and 0.6%, respectively. The optimal formulation has a particle size of 252.2 nm with a polydispersity index of 0.3828, zeta potential of -27.27 mV and a release rate of 20.0015% in 10 hours. The cycling test results of the optimal formula demonstrated good stability over a 12-day testing period.
