Abstract

The utilization of sweet orange peel is still minimal as it is often overlooked after fruit processing for consumption, despite being rich in bioactive compounds, including flavonoids, especially for topical preparations. Previous research has mainly focused on conventional formulations with limitations in skin penetration. The Nanostructured Lipid Carrier (NLC) delivery system can enhance percutaneous penetration. This study aimed to optimize the NLC formula of sweet orange peel extract based on viscosity, pH, particle size, zeta potential, mobility, and polydispersity index (PDI) characteristics. The design and optimization of the formula used central composite design with variations in the concentration of solid lipid monostearin and liquid lipid oleic acid, employing a high-speed homogenization method. The optimum formula underwent a physical stability test (cycling test) and morphology observation using TEM. The optimum formula was obtained with 3% monostearin and 11.372% oleic acid concentrations. The characteristics of the resulting optimum NLC formula included pH of 6.30±0.001, which is non-irritating to the skin, viscosity of 571.1±15.41 cps, zeta potential of -26.54±0.889 m/V, mobility of 1.1156±0.1443 mS/cm, small particle size of 367.5±1.858 nm, and PDI value of 0.3373±0.00979, indicating particle size uniformity. The physical stability results showed no changes over 6 cycles. The observed NLC morphology was spherical. Based on the obtained results, the optimum NLC formula of sweet orange peel extract demonstrated good characterization and stability.
