Abstract

Cassava leaves (Manihot Esculenta Crantz.) is proven to contain secondary metabolites, including flavonoids such as rutin, which demonstrated antibacterial capabilities. The deformability and elasticity of transethosomes penetrated the stratum corneum and increased the transfer of active substances in skin permeability. This research aimed to determine the optimum transethosome formula from variations of 96% ethanol concentration at 20% and 40%, with tween 80 concentrations of 0.5% and 0.75%, exhibiting superior antibacterial activity. Cassava leaf ethanol extract was fractionated using the liquid-liquid method with n-hexane, ethanol-water, and ethyl acetate solvents. Transethosome nanovesicles were encapsulated using the thin-layer hydration method. Optimization was performed using a 22 factorial design method with the Design Expert 12® program, focusing on the responses of encapsulation efficiency, encapsulation efficiency decline, initial pH, and pH change after stability testing. The optimum formula was characterized by particle size, PDI, zeta potential, and antibacterial activity against P. acnes bacteria. The total rutin content in the ethyl acetate fraction of cassava leaves was 33.2894 mg RE/g with a percentage of 3.32894%. Based on the Design Expert 12® analysis, the optimum transethosome formula was obtained at 40% ethanol concentration and 0.750% tween, with an encapsulation efficiency value of 82.4612±0.243, encapsulation efficiency decline of 1.469±0.174, initial pH of 7.32±0.004, and pH change of 0.04±0.008. The particle size characterization of the optimum formula revealed a particle size of 142.5±19.044 nm, PDI of 0.391±0.083, and zeta potential of -25.3±1.763 mV. The anti-acne activity testing on the optimal transethosome formula was conducted using the disc diffusion method, and a inhibition zone diameter of 14.00 ± 0.27 mm was obtained. Based on the results obtained, the optimal formula of cassava leaf ethyl acetate transethosomes exhibited good characterization and stability, and demonstrated strong inhibition of P. acnes antibacterial activity.
