Abstract

Breadfruit leaves (Artocarpus altilis (Parkinson) Fosberg) contain the main flavonoid compound in the form of quercetin which has potential as an antiosteoarthritis. Nanoemulsion preparation is carried out to increase drug stability and penetration. This research aims to determine the optimum concentration of Cremophor RH 40 and Ethanol 96% using the factorial 22. The optimum formula is determined based on pH value, density, viscosity, percent transmittance, percent total flavonoid content. The optimum formula continues with PSA, in vitro diffusion, and stability. The results showed that the optimal formula consisted of Cremophor RH 40 at a concentration of 10% and Ethanol 96% at 20%. The nanoemulsion PSA exhibited a globule size 193.566 ± 21.373 nm; PDI 0.754 ± 0,067; zeta potential -34.466 ± 0.262. In the in vitro diffusion test, the nanoemulsion preparation diffused faster than the cream preparation, indicating that the release model follows the Higuchi order. The cycling stability not experience significant changes (p<0,05) and the centrifugation test not reveal any physical changes. Overall, this study suggests that the optimized formula of the nanoemulsion can be further developed for improving the stability of breadfruit leaf extract.
