Abstract

Transdermal patch was chosen to manage the delivery system of the antibacterial compound in cassava leaves, which is rutin, that is hydrophilic. Plasticizer is one of the additional ingredients of transdermal patches that can maintain the bioavailability of a drug. Transdermal patch formulation with active cassava leaf ethyl acetate fraction ingredients and 3 variations of PEG-400 concentration as plasticizer (10%, 20%, and 30%). Transdermal patches were made using the solvent-casting method. This study aimed to determine the evaluation and stability of transdermal patch preparations based on the cycling test method and to test the antibacterial activity against the growth of Staphylococcus aureus bacteria using the paper disc method. The resulting transdermal patch preparation showed good characteristics based on organoleptic parameters, weight uniformity, thickness, percent elongation, pH, folding resistance, swelling, and humidity so the best formula was F3 with a PEG-400 concentration of 30%. The characteristics of formula 3 showed a greenish appearance, elastic and sticky; uniformity of content of 100.0005% ± 0.162; pH 4.86 ± 0.115; folding resistance >300; elongation 37.8% ± 2.020; weight uniformity 0.0617 g ± 0.001; humidity 9.16% ± 0.733; thickness 0.06mm ± 0.001 and swelling index 5.36 ± 1.245 and stable in storage for 6 cycles. The results of the antibacterial activity test showed that the transdermal patch had a strong inhibition category with a value of 10.73 ± 0.404 mm as shown by different notations against positive, standard and negative controls with a significance value of p < 0.05. Based on the results obtained, increasing the concentration of PEG-400 as a plasticizer significantly affects the characteristics of transdermal patches. The best patch formula can inhibit the growth of Staphylococcus aureus with a strong inhibitory response.
