Abstract
Ekstrak daun sukun (Artocarpus altilis (Parkinson) Fosberg) mengandung senyawa metabolit sekunder utama flavonoid dan fenolik. Penelitian ini bertujuan untuk mengetahui optimum konsentrasi Tween 80 dan Etanol 96% dalam sediaan nanoemulsi ekstrak etanol daun sukun. Optimasi dilakukan dengan desain faktorial 22 menggunakan Design Expert 12®. Formula optimum yang diperoleh selanjutnya dilakukan karakterisasi uji PSA (Particle Size Analyzer), uji Franz Diffusion Cell dan uji stabilitas dengan metode cycling test dan uji sentrifugasi. Formula optimum sediaan nanoemulsi ekstrak daun sukun diperoleh perbandingan konsentrasi Tween 80 dan Etanol 96% (40%:20%). Hasil uji PSA Formula optimum menghasilkan globul sebesar 246,1 ± 29,443 nm, PDI 0,36 ± 0,069 dan zeta potensial -12,6 ± 0 mV. Hasil uji stabilitas sentrifugasi menunjukkan tidak adanya pemisahan fase. Hasil cycling test menunjukkan penurunan pH dan persen transmitan yang tidak berbeda signifikan (p>0,05), sedangkan pada densitas terjadi penurunan yang signifikan. Formula optimum nanoemulsi terpenetrasi dengan cepat pada menit-15 sebesar 18,65% sedangkan krim 17,87%, nanoemulsi berpotensi besar menembus muscle secara in vitro dibandingkan krim. Kinetika pelepasan mengikuti orde nol dan mekanisme pelepasan model higuchi.
