Abstract
Local rice in South Sumatra is generally specific to various regions and cultivated for generations by the community and adapted to specific environmental conditions. Siam and Siputih rice survive and are planted by farmers as local varieties of swampland rice because of the good taste of rice and tolerance to swamp waterlogging for a long time, but are currently not widely cultivated because of their long life cycle and susceptibility to lodging. The introduction of early maturing superior rice varieties has caused genetic erosion in certain areas, so conservation measure are needed with a molecular DNA barcoding approach to identify an organism and maintain genetic information. The rbcL gene as a barcode in plants that have low mutation rates and high amplification success rates. This study aims to determine the sequence of different nucleotide bases of the rbcL gene of Siam rice and Siputih local varieties of South Sumatra, and reconstruct phylogenetic trees to see the kinship relationship of Siam rice and Siputih local varieties of South Sumatra. The results showed that the rbcL gene sequences of Siam and Siputih rice have high similarity indicated by significant conserved areas, but there are differences in the sequence of nucleotide bases as many as 7 points (1, 3, 589, 590, 591, 592, 593). Phylogenetic tree reconstruction of Siam and Siputih rice shows that both have a close kinship with a genetic distance of 0.000 and similarity of 100%, both also have the closest kinship with Oryza sativa cultivar Siam Mutiara with a genetic distance of 0.583 and similarity of 41.7%.
