Abstract
Changes in people's unhealthy lifestyles result in the formation of free radicals in the body. The high content of free radicals causes damage to cells and body tissues as well as metabolic disorders and causes degenerative diseases, the presence of free radicals can be prevented by antioxidant compounds. Antioxidant compounds are divided into 2 natural and synthetic antioxidants, natural antioxidants can be obtained from plants. Gelam (Melaleuca cajuputi Powell) has the potential to produce antioxidant compounds. The purpose of this study was to determine the antioxidant activity of fractions, pure compound groups and IC50 values of gelam leaves. The research methods carried out are making simplisia, extraction, fractionation, antioxidant activity test of fractions with DPPH using KLT and Spectrophotometer UV-Vis, purification and isolation of compounds, antioxidant activity test of pure compounds with DPPH using KLT and Spectrophotometer UV-Vis, and determination of IC50 value by DPPH method. The results of the antioxidant activity test of the n-hexane fraction using KLT plates obtained very strong and strong results in the ethyl acetate fraction in the water methanol fraction obtained weak results. Pure compounds obtained that belong to the terpenoid compound group are pure compounds N1, N2, N4, and E1 with sequential IC50 values of 6.00, 94.52, 33.4 and 26.72 ppm with very strong antioxidant activity, and strong. Pure compounds N6 and E6 belong to the flavonoid compound group with IC50 values of 115.64, and 50.29 ppm respectively with weak and strong antioxidant activity strength. Pure compound E3 belongs to the group of steroid compounds with IC50 value of 36.23 ppm with very strong antioxidant power.
