Abstract

ANANDA PRANSISKA. The Application of Naphtalene Acetic Acid (NAA) and Benzyl Amino Purine (BAP) on The Growth of Pontianak Sweet Orange (Citrus nobilis var. Microcarpa) Epicotyl as Explant With In-vitro Culture (Supervised by ZAIDAN and IRMAWATI). This study aimed to evaluate the effect of Pontianak Sweet Orange (Citrus nobilis var. Microcarpa) epicotyl growth at various concentrations of Naphthalene Acetic Acid (NAA) and Benzyl Amino Purine (BAP) in In-vitro culture. This research was carried out at the Tissue Culture Laboratory, Faculty of Agriculture, UNSRI located on -2.989594,104.731106. From August to October 2023. Two plant growth regulators (PGR) were used, namely NAA (N) consisted of 3 concentration levels, namely (N0=0 ppm, N0=1 ppm, N2=2 ppm) and BAP (B) had 3 levels as well, namely (B0=0 ppm, B1=1 ppm, B2=2 ppm). The results showed that NAA and BAP increased number of shoots, root formation and callus.The concentration of 1 ppm NAA and 1 ppm BAP was the best PGR combination for the growth of callus. The single treatment of 1 ppm BAP was obtained to produce the highest number of shoots and the single treatment of 2 ppm NAA was obtained to produce the highest number of roots. Microcarpa) Sebagai Eksplan Secara Kultur In-vitro (Dibimbing oleh ZAIDAN dan IRMAWATI). Penelitian ini bertujuan untuk mengetahui pengaruh pertumbuhan epikotil Jeruk Manis Pontianak (Citrus nobilis var. Microcarpa) pada berbagai konsentrasi Naphtalene Acetic Acid (NAA) dan Benzyl Amino Purine (BAP) secara kultur In- vitro. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan, Fakultas Pertanian UNSRI yang berlokasi di - 2.989594,104.731106. Pada bulan Agustus hingga Oktober 2023. Penggunaan dua zat pengatur tumbuh (ZPT) yaitu NAA (N) yang terdiri dari 3 taraf konsentrasi yaitu (N0=0 ppm, N0=1 ppm, N2=2 ppm) dan BAP (B) mempunyai 3 taraf konsentrasi juga yaitu (B0=0 ppm, B1=1 ppm, B2=2 ppm). Hasil penelitian menunjukkan bahwa NAA dan BAP dapat meningkatkan pertumbuhan jumlahtunas, pembentukan akar dan kalus. Konsentrasi 1 ppm NAA dan BAP 1 ppm merupakan kombinasi zpt terbaik untuk pembentukan kalus. Perlakuan tunggal BAP 1 ppm diperoleh dapat menghasilkan jumlah tunas terbanyak dan perlakuan tunggal NAA 2 ppm diperoleh dapat menghasilkan jumlah akar terbanyak.
