Abstract

The flavonoid content in melinjo leaves (Gnetum gnemon L.) has the potential to dissolve kidney stones through increased urinary citrate excretion, diuresis, and antioxidant effects. This study aims to examine the kidney stone dissolving activity of ethyl acetate fraction of melinjo leaves (FEADM) through urine pH, serum creatinine, ureum and kidney calcium levels, macroscopic and histopathology of rat kidneys induced by ethylene glycol and ammonium chloride. Rats were divided into 7 groups: normal control (Na-CMC 0.5%), negative control (ethylene glycol 0.75% and ammonium chloride 1%), positive control (batugin elixir 791 mg/kgBW), FEADM test group doses of 55, 110, 165, and 220 mg/kgBW. Ethylene glycol and ammonium chloride were given for 14 days followed by the administration of fractions and batugin elixir for 14 days. The results showed a decrease in mean serum creatinine and urea levels, renal calcium and an increase in urine pH value in the FEADM group doses of 55, 110, 165, and 220 mg/kgBW. LSD statistical analysis showed no significant difference between the ethyl acetate fraction of melinjo leaves at a dose of 220 mg/kgBW with the normal control group (p>0.05) on serum creatinine and ureum levels and with the positive control group (p>0.05) on kidney calcium levels. The results of macroscopic and histopathological images of the kidneys of rats in the test group also showed an improvement after FEADM administration compared to the negative control. The dose of 220 mg/kgBW provides more effective antinefrolithiasis activity than the positive control. The effective dose (ED50) of ethyl acetate fraction of melinjo leaves (Gnetum gnemon L.) was 190 mg/kgBB.
