Abstract

Keji beling (Strobilanthes crispus) is a medicinal plant that has been used traditionally in a diabetes, wound healing, diuretic, and treatment of constipation. This research aims to determine antioxidant activity the fraction of S. crispus leaves by the DPPH (2,2-diphenyl-1-picrylhydrazyl), FRAP (Ferric Reducing Antioxidant Power) dan FTC (Ferric Thiocyanate) methods. The research began with fractionation by maceration using solvents with step gradient polarities (n-hexane, ethyl acetate and methanol). Each fraction was analyzed for total phenolic, flavonoid and their antioxidant activity. The results showed that the n-hexane, ethyl acetate and methanol fractions had phenolic levels of 6.43; 11.56; 16.13 mgGAE/g, and flavonoid content of 3.75; 7.34; 7.19 mgQE/g respectively. The antioxidant activity test using the DPPH method showed that the n-hexane, ethyl acetate and methanol fractions had antioxidant activity with an each IC50 value of 731.93; 471.99; 115.69 mg/L. The antioxidant activity test using the FRAP method showed that the n-hexane, ethyl acetate and methanol fractions had antioxidant activity based on the amount of Fe2+ formed of 66.28; 138.90; 143.43 mg/L Fe2+. Meanwhile, the antioxidant activity test using the FTC method showed that the n-hexane, ethyl acetate and methanol fractions had antioxidant activity with a percent inhibition of fat peroxidation of 36.86; 55.76; 46.77% respectively. The DPPH and FRAP methods showed that the methanol fraction had higher antioxidant activity than the ethyl acetate and n-hexane fractions. These results show that antioxidant activity is proportional to the phenolic content of each fraction. Meanwhile, using the FTC method, it was found that the ethyl acetate fraction had higher activity than the methanol and n-hexane fractions. These results show that antioxidant activity is proportional to the flavonoid content of each fraction.
