Abstract

Melinjo leaves are known to have pharmacological effects as antidiabetics because they contain flavonoid compounds that are able to lower blood glucose levels by reducing glucose and regulating the excretory activity of enzymes seen in carbohydrate metabolism. Bioavailability, effectiveness, and low solubility of extracts in water are major problems in drug formulation. This can be overcome by making nanosuspension preparations. Nanosuspensions are the main choice because they can increase the aqueous solubility of drugs and bioavailability by increasing the surface area of drugs in contact with biological media. The methods used in this study are measurement of total flavonoid content, preparation of nanosuspension using an ionic gelation system, calculation of EE, and antidiabetic testing on test animals. The results of nanosuspension evaluation testing with chitosan variations of 30 mg, 150 mg, and 300 mg obtained the results of percent absorption efficiency, which are 98.416%, 95.369%, and 97.475%, respectively. Particle sizes was 162.1 nm, 156.6 nm, and 152.2 nm. PDI: 0.283, 0.287, 0.288. Zeta potential: -37.69 mV, -34.66 mV, and -31.99 mV. The results of the best formula was given to test animals that had been induced by streptozotocin and obtained the results of a decrease in glucose levels of 71.13% percent decrease in glucose levels of nanosuspension preparations, better than the percent decrease in blood glucose levels of the group given the extract, which was 65.23%, and close to the percent decrease in blood glucose levels of the positive group given metformin, which was 71.93%. The variation of chitosan in the nanosuspension preparation formula of melinjo leaf extract affects the percent of absorption efficiency and nanosuspension characteristics and shows a decrease in blood glucose levels that did not significantly different from the metformin group (p > 0.05).
