Abstract

Melinjo leaves has pharmacological activity as an antidiabetic because it contains flavonoid compounds which can inhibit glucose absorption, stimulate insulin secretion or act as insulin and can regulate the work of enzymes that play a role in carbohydrate metabolism. The effectiveness and bioavailability of extracts tend to be low so that the formula in the form of nanosuspension is an option for improving the delivery system which can increase the effectiveness of the preparation. This study aims to determine the effect of variations of sodium alginate on the characterization of nanosuspension of melinjo leaf extract and on decreasing blood glucose levels in rats with type 2 diabetes mellitus. The methods used in this research are measuring total flavonoid levels, making nanosuspension preparations using an ionic gelation system, calculating %EE and antidiabetic testing on test animals. Three formulas were used in the preparation of preparations with variations of sodium alginate, namely 0.6%, 0.3% and 0.1%. The first formula choose to be the best formula obtained an average particle size of 183,26 nm, a zeta potential of -41,92 mEv, a PDI of 0.3081 and percent efficiency of 97,4838%. The best formula was given to test animals that had been induced by streptozotocin and resulted in a glucose level reduction of 74,6814%. The percentage of glucose reduction in the nanosuspension formulation was better compared to the percentage of glucose reduction in the group provided the extract, which was 65,2378%, and approached the percentage of glucose reduction in the positive control group given metformin, which was 71,9394%. It was concluded that the variation of sodium alginate to the nanosuspension preparation formula of Melinjo leaf extract affects the percentage of adsorption efficiency and nanosuspension characteristics. In addition, it showed a decrease in blood glucose levels that did not different significantly with the metformin group (p>0,05).
