Abstract

Melinjo leaves (Gnetum gnemon L.) is a plan that is often used as a medicinal ingredient because it has a high content of secondary metabolites. Secondary metabolites that are thought to act as hepatoprotectors are flavonoid. This study aims to determine the decrease in SGPT and SGOT levels of various doses, determine ED50 (Effective Dose 50) and determine the effect on the macroscopic and histopathological of the liver treated with ethyl acetate fraction of melinjo leaves. The test animals used were divided into 7 groups, consisting of normal group that was only given a 0.5% Na CMC suspension, a positive control group using 100 mg/kgBW silymarin, a negative control groups using 0.5% Na CMC suspension and test groups I, II, III, and IV given ethyl acetate fractions of melinjo leaves at doses of 10, 20, 40, and 80 mg/kgBW, respectively. Testing was carried out for 7 days and on the 8th day all groups except the normal group were induced with CCl4 1 mL/kgBW. The parameters used were SGPT and SGOT levels as well as macroscopic and histopathological liver observations. The results of the analysis of measurement levels showed that there was a significant difference (p<0.05) between the negative control group and test groups I, II, III, and IV on SGPT and SGOT levels and there was no significant difference (p>0.05) between test group IV and positive control. Based on the relationship of the hepatoprotector percent to fraction dose, the ED50 value of the ethyl acetate fraction of melinjo leaves is 30.98 mg/kgBW. Overall, the ethyl acetate fraction of melinjo leaves can reduce SGPT and SGOT levels and provide better macroscopic and histopathological results than the negative control group, but the use of ethyl acetate fraction of melinjo leaves at a dose of 80 mg/kgBW gives the best results compared to other doses, so that the dose shows a significant preventive effect as a hepatoprotector in male white rats induced by CCl4.
