Abstract

The objective of the research was to evaluate the growth of seruni’s shoot tip explant in MS medium with NAA, BAP and coconut milk treatment. This research conducted from January to March 2008 in Plant Tissue Culture Laboratory Sriwijaya University. The treatments of this experiment were : Control (K0), Coconut milk 150 ml was 5 5 (Kl), BAP 0,5 ppm (K2), NAA 0,5 ppm (K3), NAA + BAP 0,5 ppm (K4), NAA + BAP 0,5 ppm + Coconut milk 100 ml (K5), NAA + BAP 0,5 ppm + Coconut milk 150 ml (K6), NAA + BAP 0,5 ppm + Coconut milk 200 ml (K7). The result of the reseach showed that ZPT and coconut mik treatment in medium did not to spur o the growth of seruni’s shoot tip explant to be a better. The treatment without ZPT and coconut mik (K0) gave the best result for time of shoot growth, shoot length, time of root growth, and root length. For the growth of shoot tip seruni in MS media without ZPT and coconut milk application is better, than the growth of shoot tip in media with ZPT and coconut milk application. This research indicated that seruni’s shoot tip have more endogen ZPT, so not need ZPT addition again to help the growth seruni’s shoot tip an in vitro culture.
