Abstract

Rice (Oryza sativa L.) has various rice colors based on color pigments. Red rice is a colored rice whose genetic variation is higher than other colored rice. Genetic conservation is an effort that needs to be made to save local varieties of red rice from the threat of genetic erosion and extinction. One effort to conserve the genetics of local varieties of red rice is DNA sequencing using molecular markers. DNA barcoding is one approach used to study genetic diversity. DNA barcoding is suitable for two purposes, namely molecular identification of described species and undescribed species. The gene used in this research is the rbcL gene contained in cpDNA. This gene is part of a DNA sequence located in cpDNA. The role of the rbcL gene which codes for the RuBisCO protein causes this gene sequence to have a low mutation rate compared to other barcode genes in cpDNA so that the level of similarity between species is quite high. This research aims to analyse the genetic variation of local varieties of red rice in South Sumatera. This research was carried out from May to June 2022, at the Genetics and Biotechnology Laboratory, Biology Department, Faculty of Mathematics and Science, Sriwijaya University. The methods used in this research include DNA isolation using Tiagen Plant Genomic DNA KIT DP305. Electrophoresis of the isolation results, DNA amplification using primers rbcL a-F and rbcL-724R and DNA sequencing. The research results showed that the rbcL gene in 8 samples of local South Sumateran varieties of red rice was successfully amplified with DNA bands measuring 580 bp – 588 bp and there was a high similarity with Oryza sativa cultivar accession number KP827660.1. however, there are differences in nucleotide bases between local varieties of red rice, South Sumatera, which are indicated by mutation events.
