Abstract

The objective of the research was to apply chitosanase produced by a bacterial strain JB4 isolated ffom terasi to prepare oligomers of chitosan. The research was conducted in Laboratory of Biotechnology, Research Center For Fisheries Product Processing and Biotechnology for Marine and Fisheries, Jakarta Pusat, from March 2006 to January 2007. The experiment was designed in two steps to analyze the effectivity of chitosanase to produce chitosan oligomers at different enzyme concentrations. At first step, chitosan was incubated with chitosanase (concentration of 22 U/g chitosan) at 40°C for 2, 4, 6 and 8 hours, whereas at second step chitosan was incubated with chitosanase (concentration of 5 U/g chitosan) at 40°C for 1, 2, 3 and 4 hours. Observed parameters included viscocity, composition of chitosan oligomers and their molecular weight. The results showed that chitosanase hydrolyzed chitosan effectively into chitosan oligomers, showed by reduction in molecular weight and decreasing viscocity of the samples. The dominant oligomer was trimer with viscocity between 1.03 and 1.04 cPs (first step) and 0.97 and 0.98 cPs (second step) for chitosan from prawn, whereas chitosan from crab had viscocity between 1.03 and 1.04 cPs. Chitosan oligomers originated from prawn had molecular weight of 1,349 Da and between 424 and 535 Da from the first and second steps, respectively. However, molecular weight chitosan oligomers originated from crab could not be determined.
