Abstract

Diarrhoea is an infectious disease of the human digestive system tract, one of the bacteria that can cause diarrhoeal infections is Bacillus subtilis and Bacillus cereus, natural ingredients soursop seeds (Annona muricata L.) are known to contain active compounds of secondary metabolites that are antibacterial in nature. This study was conducted to determine the ability of soursop seed extract to inhibit the growth of Bacillus subtilis and Bacillus cereus bacteria. The method used in this study is the extraction method carried out by maceration to obtain a thick extract of soursop seeds, followed by qualitative testing of bioactive compounds using reagents, isolation of active compounds by chromatography, antibacterial activity and KHM test using agar disc diffusion method. The results showed that soursop seed ethanol extract has the ability to inhibit the growth of Bacillus subtilis and Bacillus cereus, using concentrations of 6%, 5% and 4% at a concentration of 6% is the most effective concentration by producing a high enough diameter classified as strong, the results of screening bioactive compounds in soursop seeds identified 5 active compounds consisting of alkaloids, flavonoids, tannins, saponins and terpenoids, but after being isolated using column chromatography techniques, soursop seeds contain flavonoids, tannins, saponins and terpenoids. The only compounds that acted as antibacterial were terpenoids Rf 0.26 and flavonoids Rf 0.42. The minimum inhibitory concentration value obtained in Bacillus subtilis 0,5% produced an inhibition zone of 0.83 mm and Bacillus cereus 0,5% with a diameter of 1.17 mm.
