Abstract

Arcangelisia flava (yellow wood) is an Indonesian medicinal plant traditionally used to treat various diseases. Its bioactive compounds, such as phenols and flavonoids, have potential antioxidant properties. This study aimed to determine the total phenolic content, total flavonoid content, and antioxidant activity of the ethyl acetate fraction of A. flava stem. Isolation of compounds was carried out using a sequential maceration method with n-hexane, ethyl acetate, and water as solvents. Total phenolic and flavonoid content were analyzed using the Folin-Ciocalteu and quercetin methods with concentration variations of 20 ppm, 40 ppm, 60 ppm, 80 ppm, and 100 ppm, while antioxidant activity was tested using the DPPH method with concentration variations of 2 ppm, 5 ppm, 10 ppm, 15 ppm, 20 ppm, and 25 ppm, followed by IC50 value calculation. The results showed that the ethyl acetate fraction of A. flava stem contained 34.7 ± 0.74 mg GAE/g of phenols and 39.68 ± 6.34 mg QE/g of flavonoids, and exhibited strong antioxidant activity with an IC50 value of 99.73 ppm. Therefore, the ethyl acetate fraction of A. flava stem has the potential as a source of natural antioxidants to support the treatment and prevention of degenerative diseases.
