Abstract

Mycobacterium tuberculosis (Mtb) is the causative agent of tuberculosis (TB). In Indonesia, TB has a high prevalence with a total of 829.000 cases in 2023 and is the country with the second-highest number of TB cases globally. Host genetic susceptibility and environmental factors play an essential role in the mechanism of Mtb infection. Genetic polymorphism in the form of SNP rs763780 T/C in the IL-17F gene is a non-synonymous variant polymorphism that causes the substitution of histidine to arginine in position 161 (H161R), resulting in the loss of IL-17F's ability to induce the expression of specific cytokines and chemokines. This is associated with host factors in the form of an individual's susceptibility to Mtb infection. This study aims to identify the frequency distribution of the IL-17F rs763780 T/C gene polymorphism in sputum isolates from active pulmonary TB patients in Palembang. This research is a laboratory observational study to identify the frequency distribution of the IL-17F rs763780 T/C gene polymorphism from sputum DNA collections of pulmonary TB patients in Palembang. Data were obtained from the Microbiology Department, Faculty of Medicine, Sriwijaya University for the period October-November 2024 from thirty one samples that meet the inclusion criteria. This study found the heterozygous mutant genotype (TC) in 19 samples (61.29%), the homozygous mutant genotype (CC) in 1 sample (3.23%), and the wild genotype (TT) in 11 samples (35.48%). The allele frequency were 41 for T alleles (66.13%) and 21 for C alleles (33.87%). In conclusion, the mutant genotype (TC+CC) was more frequently found than the wild (TT), namely 64.52% versus 35.48%, for the allele frequency distribution, it is found that the T allele is higher than the C allele with a ratio of 66.13% versus 33.87%.
