Abstract
Acute gout arthritis is a joint inflammation in adults caused by the accumulation of monosodium urate crystals from hyperuricemia, leading to inflammation and pain. Colchicine is the primary treatment, but it has many side effects. The cherry leaf (Muntingia calabura L.), which has potential anti-inflammatory properties, offers a promising alternative treatment. This study aims to evaluate the efficacy of cherry leaf extract in maintaining interleukin-1β (IL-1β) levels in a rat model of acute gout arthritis. An in vivo experimental study was conducted over 19 days at the Animal House and Biotechnology Laboratory, Faculty of Medicine, Universitas Sriwijaya, using a pre- and post-control group design. A total of 30 Wistar rats were divided into 5 treatment groups (negative controlled, positive controlled, and EDK doses of 50, 100, and 200 mg/kgBW). Pre-induction treatment was administered for 7 days, followed by the induction of gout arthritis using 25 mg/ml MSU crystals. Data analysis was performed using SPSS version 22.0. The body weights of the test animals range between 200–300 grams and were statistically homogenous. The mean IL-1β levels in each group were 978.658±44.743; 418.133±78.164; 449.611±40.717; 460.503±37.135; and 479.058±32.562, respectively. Colchicine and cherry leaf extract were not significant in maintaining IL-1β levels (p<0.05) but effectively reduced inflammation caused by MSU crystals. The positive controlled group showed the lowest IL-1β levels compared to all EDK groups. Keywords: cherry leaf extract, interleukin-1β, IL-1β, efficacy
