Abstract
Gout arthritis is an inflammatory joint condition caused by uric acid crystal accumulation. The inflammatory mechanism involves complex pathways that increase Reactive Oxygen Species (ROS) levels. Muntingia calabura L. leaves contain bioactive compounds, such as flavonoids, which are known to reduce ROS production by mitigating pro-inflammatory signaling pathways. This study aims to evaluate the effectiveness of Muntingia calabura L. leaf extract as an anti-inflammatory and antioxidant agent in maintaining ROS levels in Wistar rats with acute gout arthritis. An in vivo experimental study was conducted using Wistar rats (Rattus norvegicus) at the Faculty of Medicine, Universitas Sriwijaya, from August to November 2024. Thirty rats were randomly divided into five groups. Four treatment groups received colchicine (0.3 mg/kgBW) or EDK (50, 100, and 200 mg/kgBW) orally for seven days. One hour after the final dose, monosodium urate (MSU) crystals were injected intra-articularly into the right knee joint. Inflammatory symptoms were assessed at 6, 24, 48, and 72 hours post-induction. ROS levels in joint tissue were measured using ELISA after 72 hours. The data were processed and analyzed using SPSS 27.0 for Windows. The 50 mg/kgBW dose of Muntingia calabura L. extract significantly maintained ROS levels compared to the negative control (Paired t-test, p<0.05). Higher doses (100 and 200 mg/kgBW) did not exhibit similar effectiveness. Muntingia calabura L. extract at 50 mg/kgBW demonstrated significant antioxidant and anti-inflammatory effects, effectively maintaining ROS levels in Wistar rats with acute gout arthritis.
